In this tutorial, I will show you how to simulate Gateway cloning with a single fragment in SnapGene.

The first step in Gateway cloning is to perform the BP reaction, which creates an entry clone with the insert flanked by attL sites. The second step in Gateway cloning is the LR reaction, which creates an expression clone where the insert is flanked by attB sites.
